FollicularregulatoryT(Tfr)cellshavespecializedrolesinmodulatingfollicularhelperT(Tfh)cellactivationofBcells.However,thepreciseroleofTfrcellsincontrollingantibodyresponsestoforeignantigensandautoantigensinvivoisstillunclear.Inthispaper,theauthorsdevelopedaTfrcell-depletedmouseinwhichtheTfrcellsweredeletedatspecifictimepointsinaninduciblemanner.TheyfoundthatthedeletionofTfrcellsbeforeGCinitiation,butnotafter,facilitatedtheGCresponsesandTfrdeletedmicehadhigherlevelsofTfhcells,Bcells.Moreover,thedeletionofTFRcellsalsoresultedinincreasedself-reactiveIgGandIgE.TheresultssuggestedthatTfrcellsregulatedearlyGCformation.ThentheyinvestigatedtheroleofTfrcellsintheHDMmodelbecauseoftheincreasedlevelofIgEandfoundthatTfrcellsregulatedIL-13productionbyTfhcellsandcontrolledIgEresponses.ThedeletionofTfrcellsinvivoduringHDMsensitizationresultedinincreasedIgEandlunginflammation.Inconclusion,thestudydemonstratedthatTfrcellswerekeyregulatorsofhumoralandallergicimmunitybycontrollingearlyGCresponses.
