Inthispaper,theauthorsmainlyfocusontheroleoffollicularregulatoryT(TFR)cellsinmodulatingfollicularhelperT(TFH)cellsandBcells.TheygeneratedtheTFR-DTR(Cxcr5IRES-LoxP-STOP-LoxP-DTRFoxP3IRES-CreYFP)micetospecificallyandeffectivelydeleteTFRcells.TheyfoundthatTFRcellspotentlyregulatedGCformationatearlystageandregulatedautoreactiveIgGandIgEantibodies.Further,theresultsledthemtoinvestigatetheroleofTFRcellsinhousedustmite(HDM)model.TheyfoundthatTFRcellsregulatedTFH13cell-mediatedIgEresponsesbothinvitroandinvivo.DeletionofTFRcellsinvivoduringHDMchallengeresultedinhigherlevelsofantigen-specificIgEandincreasedlunginflammation.TheirstudysuggestedthatTFRcellscontrolledIgGandIgEresponsestovaccines,allergensandautoantigens,andexertcriticalimmunoregulatoryfunctionsbeforeGCformation.ToexaminetheabilityofTFRcellstoregulateTFHcellsandautoreactiveBcells,theyutilizedtheTFR-TFH-Bcellsco-culturemodel,whichdeservestobestudied.

Bcellreceptor(BCR)isthecollectionofantibodiespresentinanindividualandarichsourceofpotentialtherapeuticagents.However,itssizemakesitdifficulttogetabundantinformationabouttheantigenspecificityoftheBCRsequencing.Inthispaper,theauthorsintroducedtheLIBRA-seq,themethodtomaptheantigenspecificityofBcells.Theylinkedtheoligostofluorophore-labeledantigensandsortedoutallantigen-positiveBcellsthendirectlymappedtheBCRsequenceviasingle-cellnext-generationsequencing.TodemonstratetheutilityofLIBRA-seq,theyappliedthemethodtoPBMCsamplesfromtwoHIV-infectedpatientsandidentifiedHIV-specificantibodies,includingbothknownandnovelbroadlyneutralizingantibody(bNAb)lineages.Theirmethodishigh-throughput,scalable,andapplicabletomanytargetsandmaybeatooltodiscovernewautoantigensandautoantibodiesofautoimmunediseases.
